Background: Lung cancer can be treated with surgery, chemotherapy, radiation therapy, targeted therapy, and palliative care. Palliative therapy is applied for inoperable lung cancer as it induces tumour necrosis. PH of tumour tissue is acidic; application of sodium bicarbonate (SB) into lung cancer locally via bronchoscopy can change its core pH, which may lead to tumour destruction. We aimed to study the ultrastructural characteristics of lung cancer and assess the destructive effects of sodium bicarbonate 8.4 % local injection on tumour tissue integrity by light and electron microscopies. Methods: This study was conducted on 21 patients with central bronchial carcinoma diagnosed according to WHO classification 2015. Three bronchoscopic biopsies were taken; two biopsies before and one after injection of sodium bicarbonate 8.4 % solution of 20 ml via transbronchial needle. All biopsies were examined by both light and electron microscopes. The first biopsy was examined to diagnose the tumour morphologically with and without immunostaining. Second and third biopsies were taken before and after SB 8.4 % injection to compare pathological changes in tumour tissue integrity as well as cellular ultra-structures. Different lung cancer pathological types were included in the study. Results: Tumour tissue integrity and pathological changes were examined in biopsies before and after injection of sodium bicarbonate 8.4 %. Extensive necrosis in all cell types of lung cancer was seen after injection of SB; this important finding was delineated by both light and electron microscopies. Conclusion: Preliminary ultrastructural study of small biopsy of lung tumors has a complementary role in both morphological and immunohistochemical studies. Local injection of sodium bicarbonate into lung cancer induces extensive necrosis that may reflect its important therapeutic role in lung cancer.

Huntingtin-associated protein 1 (HAP1) is abundantly expressed in the neurons of the central nervous system and forms unique intracytoplasmic inclusions of unknown function called ‘stigmoid bodies’ (STBs). Transmission electron microscopy has revealed that the STBs are aggregates of granules containing cavities with a diameter of 0.5–3 µm. Small STBs fuse to form larger STBs, the size of which is said to vary depending on the developmental growth stage and brain region. Light microscopy can only reveal that these STBs have similar circular shapes due to its limited resolution. Therefore, light microscopy is only fit for the study of the STB distribution and quantitative changes. We, herein, suggest the adoption of correlative light and electron microscopy, which combines confocal laser scanning microscopy and scanning electron microscopy as the method allowing us to identify the huntingtin-associated protein 1–positive STBs in formalin-fixed paraffin-embedded (FFPE) sections. This approach allows us to study the three-dimensional morphology of immunolabeled objects in histopathological specimens. The STBs in FFPE sections of murine hypothalami reflected the transmission electron microscopic images of Epon-embedded STBs, although we were not able to observe any organelle covering the STBs of the FFPE sections. Furthermore, we were able to reconstruct the three-dimensional structure of the STB, and we identified it to be of spherical form, covered with mitochondria and rough endoplasmic reticulum, and bearing a cluster of cavities in the centre. In the future, we might gain new insights by comparing the three-dimensional structure of the STB between different neurons and under a variety of conditions.

Introduced by Cowles Communications and Visual Panographics in 1964, the xograph® or parallax panoramagram, was the first lenticular, autostereoscopic, photomechanical object created for the mass media. Publications such as LOOK magazine and Venture: A Traveler’s Guide frequently distributed xographs® during the 1960s and 1970s, after which time, the xograph® began to disappear from mass publications. The thesis provides a detailed account of the history of three-dimensional photographic techniques and places the xograph® within this history. It addresses the contributions and collaboration of Arthur Rothstein, Marvin Whatmore, Visual Panographics and Cowles Communications in the creation, production and dissemination of xographs®. The thesis then describes xograph® production process and the results of an electron microscopic analysis of an xograph® made to determine its physical properties . The conclusion offers suggestions for preservation guidelines for these fascinating objects.

We performed histological and electron microscopic analysis of miracidia of Schistosoma mansoni in order to examine their germinal elements. In total, about 20 germinal cells at different stages of maturation were found. We described their ultrastructure and proposed a scheme of reproduction of mother sporocysts of S. mansoni based on our data and literature information. According to this scheme, the only germinal elements present in the miracidia are germinal cells (undifferentiated cells were not found). Regardless of their size and localisation, none of the germinal cells in the miracidia has undergone full differentiation. This process is completed after the metamorphosis of the larva into the sporocyst.

Background and Aim: Pediculus humanus capitis, the human head louse, remains a global health problem. This study evaluated the resistance of head lice to permethrin and 6-paradol mediated by in vitro detoxification enzyme activity experiments and to describe physical changes in the lice using scanning electron microscopy (SEM). Materials and Methods: The adult stages of P. h. capitis were collected from patients exposed to 1% permethrin and three different concentrations of 6-paradol (0.00005%, 0.0001%, and 0.00015%) using a filter paper diffusion bioassay. Healthy P. h. capitis adults served as the control. The in vitro bioassays were conducted after 10, 20, 30, and 60 min of exposure. The activities of acetylcholinesterase (AChE), glutathione S-transferase (GST), and oxidase were analyzed. Physical changes in the lice were analyzed using SEM. Results: Permethrin and 6-paradol exhibited low toxicity against the lice. At 60 min, 1% permethrin had killed 36.7% of the lice present, while 6-paradol had killed 66.7-86.7%. Permethrin induced significantly elevated AChE, GST, and oxidase activity; 6-paradol also caused significantly elevated AChE, GST, and oxidase activity. Permethrin did not cause any ultrastructural morphological changes on the lice, while 6-paradol severely damaged the head, thorax, respiratory spiracles, and abdomen of the dead lice. Conclusion: This in vitro experimental of P. h. capitis is the first study to report P. h. capitis in East Jakarta shows complete resistance to permethrin and 6-paradol, and to describe the associated increase in AChE, GST, and oxidase activity. It was observed that 6-paradol severely damaged the head, thorax, respiratory spiracles, and abdomen of the dead lice.

Pollen is appropriately referred by some as Golden dust extremely valuable on account of their tremendous applications in science, industries and public health. No other plant part even though extremely tiny in size is packed with so much information and power. Similar to other plant parts, pollen characters are so varied that the classification system of plants can be built up entirely on the basis of pollen morphology.Palynology is the distinct branch of biology that deals with the dispersed microscopic tiny living and fossil entities including pollen grains, spores, algal and fungal fragments and others. An important aspect of Palynology is the Pollen morphology. The importance of Palynology in taxonomic and phylogenetic consideration of plants is well known. The changes occurring through hybridization and years of cultivation are reflected in pollen morphology. The scope and interest in the study of pollen morphology have widened with the advent of Scanning Electron Microscopy (SEM) and with regards to unipalynous taxa particularly the understanding of finer morphology is of fundamental importance. SEM gives a correct understanding of exine surface as the electron photographs of the surface replica of the exine provides the exact picture of the ornamentation pattern. The variation in the pollen morphological characters helps in the classification of plant taxa and their assessment of their phylogenetic relationship. In the present investigation, the pollen morphological studies were carried out of some ethnomedicinal plants like Catharanthus roseus, Allamanda cathartica, Datura metel, Brassica juncea, Raphanus sativus and Cleome viscosa pollen grains by Scanning electron microscopy. All that they possess anticancer characteristics in common.

The aim of the study was to study the characteristic features of ultrastructural changes in the cellular and extracellular matrix of the adrenal and thyroid glands in chronic hypoxia.Materials and methods. The study used the thyroid and adrenal glands of healthy adult male white rats weighing 180-200 g. The anatomical, histological, histochemical, electron microscopic and morphometric methods were used.Results. We found that the response of thyroid and adrenal cells to hypoxia is different. So, since the processes of proliferation in the cells of the thyroid gland occur faster and earlier, all tissues are restored on the 15th day of the experiment in the thyroid gland, and in the adrenal gland are restored only on the 30th day of the experiment. And this can be regarded as a higher degree of sensitivity of the adrenal glands to hypoxia, which is a stronger stress factor than the thyroid gland.Summary. The structures of the thyroid gland adapt to long-term hypoxia earlier, and responds to this with ultrastructural rearrangement – hyperplasia, hypertrophy and proliferation of thyrocytes.

The paraepiglottic tonsil present on either side of the base of the epiglottis constitutes a small portion of the mucosa-associated lymphoid tissue. The present study was conducted on the paraepiglottic tonsils of 12 adult buffaloes of the local mixed breed to explore its basic histomorphological, histochemical, and ultrastructural characteristics and to compare with other tonsils of the oral region to ascribe immunological function. The tonsil mucosa was lined by partly stratified squamous keratinised epithelium towards the outer surface and a stratified squamous non-keratinised epithelium towards the shallow crypts. The non-keratinised epithelium modified into reticular type due to infiltration of underlying lymphoid tissue was characterised by the predominance of the lymphoid cells, interrupting basement membrane and indistinct strata. The reticular epithelium at places presented a spongy appearance. The loose irregular connective tissue of propria-submucosa was mainly comprised of glandular and lymphoid tissue. The lymphoid tissue was mainly in the form of isolated lymphocytes, diffuse aggregations, and lymphoid follicles. The inter-follicular areas showed the presence of high endothelial venules. The sero-mucous type of glandular acini presented a strong reaction for glycogen, acidic mucosubstances, weakly sulfated mucosubstances, mucin, and the presence of more than 4% cysteine in their secretions. The tonsils under scanning electron microscopy presented an undulating appearance due to the presence of crest and folds. Higher magnification revealed squamous cells having various patterns and arrangements of microplicae. Small pits visualised on the mucosal surface represented openings of the glandular ducts. The cells towards the lumen of some glandular ducts appeared whorl-like where typical microplicae were absent. The transmission electron microscopy of different strata of different types of epithelia showed varying cell organelles, especially the mitochondria, endoplasmic reticulum, Golgi apparatus, and few filaments. The structural features of the tonsil were similar to those of other oral tonsils and suggested their immunological role, which might be of importance to local immunity.

Chemotherapeutic agent induces severe tissue injury. Royal jelly has beneficial biological effects. Aim: To evaluate the protective role of royal jelly on salivary glands of rats treated with cytarabine. Methods: 21 male albino rats were divided into three groups. Group I (control group) normal without any medication. Group II (cytarabine group) at dose 100mg/kg/ I.P. Group III (cytarabine & Royal jelly group) was administrated royal jelly prior to cytarabine injection. After 2weeks the submandibular glands were dissected and prepared for histological and electron microscopic (TEM) examinations. Results: Light microscopic examination of cytarabine group revealed deformity of the secretory portions with numerous intracellular vacuoles, deeply stained atrophied nuclei. Dilated excretory ducts with degenerated epithelial lining. Some secretory cells were degenerated. While Royal jelly treated group revealed well defined serous acini and well form striated ducts. TEM examination of cytarabine group revealed pyknotic nucli with nuclear polymorphism. Swollen mitochondria and dilated RER. Striated duct has ill-defined cell boundaries and cytoplasmic vacuolizations. These changes were improved at Royal jelly treated group. Conclusion: Administration of Royal jelly produced a protective effect against cytotoxic changes induced by chemotherapy in rat submandibular salivary glands.

PURPOSE: To describe and compare the histological changes in the cut edges of the remaining donor corneal rim using femtosecond laser-assisted keratoplasty (FAK) versus conventional penetrating keratoplasty (PK) via light and transmission electron microscopic examination. METHODS: This was a prospective observational study of 10 eyes; 5 FAK (top-hat technique) and 5 conventional PK. Main outcomes were histological findings at the cut edge of the donor corneal rim (at 3, 6, 9, and 12 o'clock). RESULTS: Cellular and ultra-cellular changes in the form of stromal edema, disorganized collagen fibers, and nuclear changes were more prominent in the FAK eyes as compared to the conventional PK ones. CONCLUSION: FAK induces more collateral damage in the cut edge of corneal donor graft at cellular and ultra-cellular levels, compared to conventional trephination. Further studies are required to investigate the clinical ramifications of this observation.

Detrimental to the performance of solid oxide cells (SOCs) is the interface between electrolyte and each electrode: for purely electron-conducting electrodes, it is part of the triple phase boundary where the reactions take place, and for mixed ionic and electronic conductor (MIEC) electrodes, the oxygen ions coming from or moving into the electrolyte are transported through this phase boundary. As such, the performance of SOFCs and SOECs is strongly dependent on the chemical nature of this interface. By combining state-of-the-art electron microscopy, synchrotron-based X-ray spectroscopy and theoretical calculations, we were able to obtain an atomically resolved picture of the interface between yttria-stabilized zirconia (YSZ, with 8 mol% Y2O3) and lanthanum strontium manganite (LSM, (La0.8Sr0.2)0.95MnO3-δ).[1] Energy-dispersive X-ray (EDX) spectroscopy measurements in the transmission electron microscope (TEM) reveal the presence of a significant amount of inter-diffusion between YSZ and LSM. Only strontium does not show any diffusion into YSZ, resulting in a shift of its intensity distribution profile of approximately 0.8 nm with respect to all other elements. All this is in agreement with Monte-Carlo-based simulations that were used to theoretically model the YSZ/LSM boundary including the ion- as well as site-specific swapping probabilities for the diffusion process.[1] According to these simulations, this diffusion region is slightly amorphous. By means of atomically resolved scanning TEM (STEM), this reduction of long-range order was observed experimentally for an approximately 1.5 nm wide slab on the YSZ side of the boundary, clearly distinguishing it from the bulk fluorite structure. In materials science, distinct ‘2D-like’ layers at grain boundaries and interfaces are often referred to as ’complexions’. While these can in general occur in different types, depending on thickness and retained order,[2] they are all characterized by a self-limited (finite) width and thermodynamic stability obtained only by confinement in between two bulk phases (i.e. they cannot exist on their own without neighboring phases).[2] These complexions have recently also been discovered in other energy-related systems such as battery materials.[3] The presence of such a stable complexion might be the reason why the YSZ/LSM interface is not as prone to the formation of lanthanum or strontium zirconates as other perovskites such as lanthanum strontium cobalt ferrite (LSCF).[4] Consequently, it might have significant influence on the chemical stability of such SOCs and a thorough understanding of the complexions between electrolyte and electrode may in the future allow fine tuning of SOC performance and stability. This requires the determination of electrochemical and thermodynamic properties of these interface complexions for which we will, in a next step, employ theoretical simulations and experimental techniques in order to find out how different transport phenomena, widths and stabilities of the layers behave with temperature, time and chemical environment. Nucleobases are nitrogenous bases composed of monomers that are a major constituent of RNA and DNA, which are an essential part of any cellular life on the Earth. The search for nucleobases in the interstellar medium remains a major challenge, however, the recent detection of nucleobases in meteorite samples and laboratory synthesis in simulated analogue experiments have confirmed their abiotic origin and a possible route for their delivery to the Earth. Nevertheless, cellular life is based on the interacting network of complex structures, and there is substantial lack of information on the possible routes by which such ordered structures may be formed in the prebiotic environment. In the current study, we present the evidence for the synthesis of complex structures due to shock processing of nucleobases. The nucleobases were subjected to the reflected shock temperature of 3500-7000 K (estimated) and pressure of about 15-34 bar for over �2 ms timescale. Under such extreme thermodynamic conditions, the nucleobases sample experiences superheating and subsequent cooling. Electron microscopic studies of shock processed residue show that nucleobases result in spontaneous formation of complex structures when subjected to extreme conditions of shock. These results suggest that impact shock processes might have contributed to the self-assembly of biologically relevant structures and the origin of life. Copyright © The Author(s), 2021. As a result of an increase in vascular permeability under the influence of intravenously administered endotoxin (LPS) in subcapsular, perivascular, perineuronal and interneuronalspaces of the spinal ganglia were identified unstained areas corresponding to theplacement of edematous fluid. Dimensions of the "perineural windows" formed in the inner layerspinal ganglion capsule depends on the number and perimeter (exchange surface area)vessels of the spinal ganglia. Disruption of the relationship between pseudo-unipolar neuronsand glial satellite cells leads to the formation of dark degenerative neurons.It was ultrastructurally proved that the cause of pyroptosis death of macrophages located insideIn the spinal ganglia, it is not the E. coli itself, but the LPS, which is part of its wall.

Conducted electron microscopic investigation of the primary remex fine structure of thirteen species of Owls (Strigiformes), using a scanning electron microscope (SEM). It is shown that Owls (Strigiformes) have a number of specific primary remex microstructural characteristics. First of all, these are the features of the structure of the pennaceous barb: a cross section configuration, a pith architectonics on the cross section and longitudinal sections, a cuticular structur of the barb. A number of the unique features in the microstructure of the vanules of the pennaceous barb have been found for the first time (at the scanning electron microscope level, at a large SEM magnification). First of all, these are the structural features of the distal barbules and the structure of the apical portion of the barb with the elongated proximal barbules and the distal barbules tightly contiguous to the ramus and closed with each other. Mentioned characteristics make for the thick velvet-like dorsal surface of the vane and the presence of a complex of peculiar “bunches” (fringes) forming the cleft edge (a fringed edge) of the inner vane – exceptionally specific adaptive characteristics in Strigiformes. Рresentenced original research results suggest that Owls (Strigiformes) have a number specific microstructural characteristics of the primary remex and also a number of the unique features in the microstructure of the primary remex which reflecting the ecological and morphological adaptations conditioned by the flight specificity.

The discovery of Langerhans and microscopic description of islets in the pancreas were crucial steps in the discovery of insulin. Over the past 150 years, many discoveries in islet biology and type 1 diabetes have been made using powerful microscopic techniques. In the past decade, combination of new probes, animal and tissue models, application of new biosensors and automation of light and electron microscopic methods and other (sub)cellular imaging modalities have proven their potential in understanding the beta cell under (patho)physiological conditions. The imaging evolution, from fluorescent jellyfish to real‐time intravital functional imaging, the revolution in automation and data handling and the increased resolving power of analytical imaging techniques are now converging. Here, we review innovative approaches that address islet biology from new angles by studying cells and molecules at high spatiotemporal resolution and in live models. Broad implementation of these cellular imaging techniques will shed new light on cause/consequence of (mal)function in islets of Langerhans in the years to come., Innovative microscopic approaches that allow to address islet biology from new angles by studying cells and molecules at high spatiotemporal resolution and in live models are reviewed in this article. Broad implementation of these cellular imaging techniques will shed new light into the cause/consequence of (mal)function of islet of Langerhans in the years to come.

Dragon fruit (Hylocereus spp.) is an important fruit crop for its high nutrition and profitability grown in India and many other countries. Dragon fruit stem samples exhibiting chlorosis and mottling symptoms, collected from Sangareddy district of Telangana, India revealed a flexuous rod-shaped virus with a length of ~ 600 nm in electron microscopic examination. The RT-PCR based detection of Cactus virus X using specific primers resulted in the production of ~ 900 bp amplicons covering the full length triple gene block protein 3 and coat protein regions. Further, cloning and sequence information of the ~ 900 bp amplicon revealed the presence of Cactus virus X (CVX) in dragon fruit. To our knowledge, this is the first record of CVX causing particular disease in dragon fruit from India.

The aim of this study is to study the effect of dark deprivation on the ultrastructure of epidermal keratinocytes. Electron microscopic evaluation of the status of keratinocytes of the epidermis of the skin of white outbred rats with dark deprivation. For morphometric evaluation of the production capacity of counting the number of granules per 100 μm2. Using the application program ImageScopeM determines the average and equivalent indicators of lamellar bodies (μm), as well as their perimeter (μm); the average area of one section of the lalellar body (μm2); roundness coefficient and average relative electronic number of lamellar bodies.Electron microscopic analysis of keratinocytes revealed significant changes in their ultrastructure, which are due to the influence of desynchronosis. Maximum transformations were observed in Odland granules (lamellar bodies). Thus, the morphometric assessment revealed changes in the number (increase), their size (decrease), as well as in the distribution of lamellar plaques in them.With desynchronosis, significant changes in the ultrastructure of keratinocytes are observed. This indicates significant systemic disorders of the epidermis as a whole. In this transformation, there are cells of a special prickly and granular layer, in particular, lamellar bodies.

Candida species are opportunistic fungi that can cause mucosal or invasive infections. Especially in biofilm-related infections, resistance is very high to anifungals; therefore more effective treatment strategies are needed. Farnesol(3,7,11-trimethyl-2,6,10-dodecatriene-1-ol) is the quorum sensing (QS) signal molecule and can interact with Candida species both as a QS molecule and as an exogenous agent. The aim of this study was to investigate the effects of farnesol on both the planktonic and biofilm forms of Candida species by colorimetric, microbiological, and electron microscopic methods. Obtained results demonstrated the inhibitory effect of farnesol on the planktonic and biofilm forms of Candida. Farnesol showed a biofilm-enhancing effect at lower concentrations. TEM findings showed the membrane and wall damage, vacuolization, or granulation in cells. SEM images confirmed biofilm reduction in pre-/post-biofilm applications as a result of farnesol treatment. In conclusion, farnesol can be used as an alternative agent to reduce the Candida biofilms, with future studies.

This work presents the experimental data obtained using an integrated approach in the study of the chemical, crystallographic and morphological homogeneity of the structure of a heat-resistant material on a nickel base with carbide-intermetallic hardening and an increased content of the γ-phase, synthesized on single-crystal substrates of various compositions with CHO in Z-direction. Using the proposed method for studying the capabilities and certification of different-level structural structures of samples in the initial state and after thermal study of the impact. The analysis was carried out using the system for EBSD analysis integrated into the scanning electron microscope and the software package for the analysis of electron microscopic images.

The characteristic mechanical activities of the smooth muscles found in all organs of the body are highly variable and depend mainly on the spatial arrangement of the muscle cells and the stroma: mass, orientation, relationships, links, constraints, which are deployed in various configurations. These structural features are examined here for their mechanical relevance, in light and electron microscopic views of several muscles of viscera and blood vessels, in a selection of mammalian species. Smooth muscles are incompressible and therefore maintain constant volume. They do not have available space and any movement of a part requires displacement of another part. Most of them have no terminations or points of attachment, and in hollow organs such as intestines, blood vessels and uro-genital tract they usually form structures closed onto themselves, such as rings or bag-like containers In these situations, changes in the size of the lumen is achieved very efficiently by a concentric inward enlargement that accompanies muscle contraction. The longitudinal arrangement of collagen blocks an elongation of small blood vessels upon contraction, further enhancing the efficiency of lumen reduction. In other muscles, links between layers and special arrangements of the stroma allow both shortening and elongation of a tubular organ to occur. The mechanics of smooth muscles has many characteristic features (some unique, some shared with those of hydrostats, some at variance with other muscles) and histological data are a contribution to our understanding of these properties.

The article presents the results of electron microscopic investigations of populations of pathogenic Candida albicans № 138 from the museum collection. The morphology of the fungus, its ultrastructure and phases of development in the population were studied. The methods developed at VNIIVSGE for growing bacteria and fungi on the surfaces of Millipore membrane filters without disturbing the architectonics of populations were used in research. The preparations were examined with electron microscope Hitachi 800 (Japan) with two systems: transmission and scanning. At large magnifications, the features of the ultrastructure of Candida albicans cells were studied. It was found that in addition to budding, Candida albicans has a survival strategy in the form of the formation of small blastospore cells, the structure of which is visible only with the use of a scanning electron microscope. The forms, sizes and localization of blastospores with their own development cycle have been studied. The morphology of pseudogyphs, which play an important role in the existence and survival of fungi population, has been studied. The formation of biofilms at a certain stage of development of Candida albicans population was revealed.

Comparative analysis of age-related changes in the ultrastructure of the mitochondrial apparatus of skeletal muscle was performed using electron microscopic and morphometric methods in specimens with different aging programs: short-lived, well-studied classical species -mice (C57BL \ 6), Wistar rats; prematurely aging OXYS rats; and animals with delayed aging - naked mole-rat (Heterocephalus glaber). In mice age-related changes in organization of the mitochondrial apparatus of skeletal muscle was shown to correspond with the previously studied for Wistar rats: the mitochondrial reticulum is formed to 2.5-3 months of age, to 30 months of age there is a significant reduction of the mitochondrial apparatus as the result of a decline in the number of mitochondria – number of sections of mitochondria in muscle fibers is reduced by almost 2 times – from 0.45±0,074 sections per µm2 to 0.23±0,017 sections per µm2. Destructive changes in the mitochondrial ultrastructure were not observed, in contrast to OXYS rats, in which age-related changes in the chondriome affect both the overall structure of the mitochondrial apparatus of muscle fibers and the internal ultrastructure of organelles. At the same time, in a small rodent similar in size to mice, Heterocephalus glaber, the number and size of mitochondria in skeletal muscle increases significantly by the age of five, but the mitochondrial reticulum does not form.

In the historical aspect, experimental studies on rabbits conducted after the discovery of penicillin are described in order to develop single doses of the antibiotic and the frequency of its administration. The effectiveness of specific therapy of patients with syphilis with crystalline penicillin, Durant drugs in monotherapy, in combination with bismuth and non-specific therapy was evaluated. Electron microscopic studies that established the mechanism of action of penicillin on Treponema, the appearance of cysts and L-forms of the pathogen, the role of the macroorganism and stimulating therapy in the completion of phagocytosis are highlighted. It reflects the discussions of leading scientists at congresses that contribute to the development of effective treatment regimens for patients with syphilis.

Thymosin β4 (Tβ4) was extracted forty years ago 1 from calf thymus. Since then, it has been identified as a G-actin binding protein involved in blood clothing, tissue regeneration, angiogenesis, and anti-inflammatory processes. Tβ4 has also been implicated in tumor metastasis and neurodegeneration. However, the precise roles and mechanism(s) of action of Tβ4 in these processes remain largely unknown, with the binding of the G-actin protein being insufficient to explain these multi-actions. Here we identify for the first time the important part of Tβ4 mechanism in ferroptosis, an iron-dependent form of cell death, which leads to neurodegeneration and somehow protects cancer cells against cell death. Specifically, we demonstrate four iron2+and iron3+ binding regions along the peptide and show that the presence of Tβ4 in cell growing medium inhibits erastin and glutamate-induced ferroptosis in macrophage cell line. Moreover, Tβ4 increases the expression of oxidative stress-related genes, namely BAX, hem oxygenase-1, Heat shock protein 70 and Thioredoxin reductase 1, which are downregulated during ferroptosis. We state the hypothesis that Tβ4 is an endogenous iron chelator and take part of iron homeostasis in ferroptosis process. We discuss the literature data of parallel involvement of Tβ4 and ferroptosis in different human pathologies, mainly cancer and neurodegeneration. Our findings confronted with literature data shows that controlled Tβ4 release could command on/off switching of ferroptosis, and may provide novel therapeutic opportunities in pathologies of cancer and tissue degeneration.

Black phosphorus is one of the emerging members of two-dimensional (2D) materials which has recently entered the biomedical field. Its anisotropic properties and infrared bandgap have enabled researchers to discover its applicability in several fields including optoelectronics, 3D printing, bioimaging, and others. Characterization techniques such as Raman spectroscopy have revealed the structural information of Black phosphorus (BP) along with its fundamental properties, such as the behavior of its photons and electrons. The present review provides an overview of synthetic approaches and properties of BP, in addition to a detailed discussion about various types of surface modifications available for overcoming the stability-related drawbacks and for imparting targeting ability to synthesized nanoplatforms. The review further gives an overview of multiple characterization techniques such as spectroscopic, thermal, optical, and electron microscopic techniques for providing an insight into its fundamental properties. These characterization techniques are not only important for the analysis of the synthesized BP but also play a vital role in assessing the doping as well as the structural integrity of BP-based nanocomposites. The potential role of BP and BP-based nanocomposites for biomedical applications specifically, in the fields of drug delivery, 3D printing, and wound dressing, have been discussed in detail to provide an insight into the multifunctional role of BP-based nanoplatforms for the management of various diseases, including cancer therapy. The review further sheds light on the role of BP-based 2D platforms such as BP nanosheets along with BP-based 0D platforms—i.e., BP quantum dots in the field of therapy and bioimaging of cancer using techniques such as photoacoustic imaging and fluorescence imaging. Although the review inculcates the multimodal therapeutic as well as imaging role of BP, there is still research going on in this field which will help in the development of BP-based theranostic platforms not only for cancer therapy, but various other diseases.

Objective – to determine the micro- and ultramicroscopic features of articular cartilage of the knee joint in rats under conditions of multiple skeletal trauma of the femoral and tibial diaphysis.Material and methods. The experiment was performed on 40 white laboratory mature male rats, divided into 2 groups: І – control group (20 rats); ІІ - study group (20 rats). Group II rats were simultaneously simulated the holey defect in the femoral and tibial diaphysis. The study of the articular surfaces of both bones was performed 6 months after the injury. Light microscopy was performed using an Olympus BH-2 microscope (Japan), Ultramicroscopic examination was performed using a transmission electron microscope (JEM-1230, JEOL, Japan) and a scanning electron microscope "SEO-SEM Inspect S50-B".Results. An articular cartilage of the proximal tibial epiphysis was the most vulnerable under conditions of multiple skeletal trauma of the femoral and tibial diaphyses. Moreover, the destructive transformations have developed in all its zones. The deep zone was almost completely replaced by bone tissue with remnants of chondrocytes. In the articular cartilage of the distal epiphysis of the femur, the destructive changes and destruction (death) of chondrocytes also occurred but to a lesser extent. The articular surface was more remain, there were no destructive changes in comparison with the articular surface of the proximal epiphysis of the tibia. The intermediate and deep zone was well traced but contained chondrocytes which were in different functional states.Conclusions. The identified structural changes in the articular cartilage of the knee joint indicate the high risk of progression of articular cartilage degradation and the development of osteoarthritis.

The syrinx is the main source for phonation in birds, its function is analogous to the mammalian larynx. Birds have both a larynx and a syrinx, but they use only the latter to vocalize. The objective of this work to give a detailed description of the anatomical, histological, and ultrastructural of syrinx in male budgerigars as a model of a passerine bird. The syrinx in the current study was to be found as a tracheobronchial type, it consists of cranial (tympanum) part and caudal (bronchosyringeal) part and, additionally, there are lateral vibrating membranes. The tympanum is formed of the last six tracheal rings, histologically its lamina epithelialis is a pseudostratified ciliated columnar epithelium with goblet cells and interrupted by intraepithelial glands. The secretory acini appear oval and lined by pyramidal secretory cells. The lamina propria–submucosa contain numerous blood capillaries, immune cells, and telocytes (TCs). The electron microscopic examination revealed numerous blood capillaries surrounded by fibroblasts and numerous immune cells, including mast cells and wandering leukocytes, within the tympanum mucosa. Hence, this study provides a detailed knowledge about the syrinx in male budgerigars.

Background: Monosodium Glutamate (MSG) is one of the most widely used food-additives in commercial foods. The excessive MSG administration may lead to damage of many organs including the testes. Ginger is medicinal plant, medically used for its antioxidant protection activity and it has a beneficial effect on male reproductive functions. Aim of the study: This study was performed to find out the effect of MSG on the histology of the seminiferous tubules of adult albino rats and possibility of recovery without treatment and administration of ginger. Material and methods: sixty adult male albino rats were divided into three equal main groups, each of which included 20 rats. Control group (C) kept without treatment, treated group (T) which divided into two subgroups (T1): which were given monosodium glutamate daily orally for 14 days. (T2): which were given monosodium glutamate daily orally and ginger daily orally for 14 days. Recovery group (R) which divided into two subgroups, (R1): which subdivided into two subgroups:(R1a) & (R1b): which were given monosodium glutamate daily for 14 days followed by 6and 13 days without treatment sequentially. (R2): subdivided into two subgroups:(R2a) & (R2b): which were given monosodium glutamate daily and ginger for 14 days followed by continuous treatment with ginger only for 6 and 13days sequentially. Results: MSG causing alteration in the histological of testicular tissues. The testicular damage caused by MSG is reversible without treatment or with administration of Ginger. Conclusion: Ginger has protective effects against MSG on the rat testicular tissue.

Objectives: to evaluate the effect of milled teeth versus beta tricalcium phosphate bone graft material on new bone formation at the cell level using transmission electron microscope Materials and methods: Forty-five adult male albino rats, were divided into three main groups (n=15); control and two experimental groups. Under general anesthesia, 1.5 cm incision was done exposing the cranial bone. Then two noncritical size bone defects were created one in each parital sides of the rat calvarial bone. In Group I (control group): the bilateral calvarial bony defects were left to heal spontaneously. In Group II the bilateral calvarial bony defects completely filled with β-TCP. In Group III: the bilateral calvarial bony defects completely filled with tooth ash. Five rats from each group were sacrificed after 7, 15, and 30 days postoperatively. Results: tooth ash group: showed more new bone formation. Different types of cells appeared more active: multiple lymphocytes, fibroblast, osteoprogenitor cells, osteoblasts. Multinucleated osteoclasts with prominent vacuolated cytoplasm and ruffled border in Howship’s lacunae were clearly seen. At day 30, disappearing of the tooth ash and infiltration of different types of cells; osteoprogenitor cells, bone lining cells, and osteoblasts while β-TCP particles were still seen in the sections. Conclusion: Bone activity, formation and maturity were ahead in tooth ash filled calvarial bony defects. Tooth ash had a role in the stimulation of osteoblastic activity in matrix formation and its mineralization.

Different types of biological grafts are useful for the replacement of infected prostheses in vascular surgery. In cases, when autologous grafts or homografts are not available, xenografts can be an alternative choice. To reduce immunogenicity, decellularization of the xenogeneic graft is necessary. Our goal was to determine the long-lasting stability of our decellularized porcine vascular grafts, in the mirror of differential scanning calorimetry (DSC) and routine histological examination. In the first experiment, light and electron microscopic confirmation of adequate decellularization of our used protocol was assessed. In the second series, DSC measurements were performed both on native and on treated samples; thereafter, histological evaluation of the analyzed samples was performed. Light and electron microscopy evaluation showed complete removal of cells and preserved connective tissue fiber structure after decellularization in the first series. In the second part of our investigation, DSC measurements demonstrated reversible change in the structure of elastic fibers and irreversible change in the structure of collagen fiber structure supported by histological findings. Our examination strengthens the earlier observations that decellularized biological grafts can tend to late aneurysmatic degeneration, due to their weakened fiber structure, besides the known immunological circumstances.

One of the important tasks of modern science is to find biochemical markers that would be able to reflect the risks of development and the nature of the course of various diseases, as well as to predict their possible consequences. In recent years, a significant number of compounds that can affect the biochemical profile of the organism have been identified. Homocysteine – a product of methionine metabolism, belongs to one of these markers, and the effects of its influece on the structure and function of various organs are being actively studied by modern researchers. The aim of the study is to find the patterns of electron microscopic changes in the liver structure of adult rats with hyperhomocysteinemia. The experimental study was performed on 22 white nonlinear mature male rats, which were divided into a control group and an experimental group. A model of persistent hyperhomocysteinemia was created by administering to rats of experimental group thiolactone homocysteine at a dose of 200 mg/kg body weight intragastrically for 60 days. The study of ultrastructural changes in the liver of rats was performed using an electron microscope PEM-125K. In adult rats with experimental hyperhomocysteinemia at the ultrastructural level, dystrophic and destructive changes in hepatocytes, endotheliocytes in the walls of sinusoids and Kupffer cells were found. These changes were more pronounced than in young rats with experimental hyperhomocysteinemia. Revealed structural changes in decompensation (depletion) of mitochondria – fewer number of cristae and enlightened matrix. In contrast to young rats, adult rats with hyperhomocysteinemia in the perisinusoidal spaces showed elongated Ito cells, a significant proportion of the cytoplasm is occupied by the Golgi complex and granular endoplasmic reticulum tanks, indicating protein synthesis for export. In Ito cells, the content of fat droplets, which are located on opposite poles of cells, is reduced. This morphological picture manifests the transformation of Ito cells into fibroblasts.

The results of determining the porosity of chemical nickel coatings, which were obtained using electron microscopy and an image processing system, are reported. The procedure allows pores more than 10 nm in diameter to be detected. Data on changing the porosity of chemical nickel coatings subjected to full-scale tests in a sea atmosphere are obtained. A three-stage failure mechanism of cathode coatings is suggested. The service life of coatings is determined.

Granum is a basic structural unit of the thylakoid membrane network of plant chloroplasts. It is composed of multiple flattened membranes forming a stacked arrangement of a cylindrical shape. Grana membranes are composed of lipids and tightly packed pigment-protein complexes whose primary role is the catalysis of photosynthetic light reactions. These membranes are highly dynamic structures capable of adapting to changing environmental conditions by fine-tuning photochemical efficiency, manifested by the structural reorganization of grana stacks. Due to a nanometer length scale of the structural granum features, the application of high-resolution electron microscopic techniques is essential for a detailed analysis of the granum architecture. This mini-review overviews recent approaches to quantitative grana structure analyses from electron microscopy data, highlighting the basic manual measurements and semi-automated workflows. We outline and define structural parameters used by different authors, for instance, granum height and diameter, thylakoid thickness, end-membrane length, Stacking Repeat Distance, and Granum Lateral Irregularity. This article also presents insights into efficient and effective measurements of grana stacks visualized on 2D micrographs. The information on how to correctly interpret obtained data, taking into account the 3D nature of grana stacks projected onto 2D space of electron micrograph, is also given. Grana ultrastructural observations reveal key features of this intriguing membrane arrangement, broadening our knowledge of the thylakoid network’s remarkable plasticity.

A large part of the Antarctic surface is covered by lichens since they can withstand extreme environmental conditions. Lichens are primary colonizers and contribute to soil formation by deteriorating rocks through a combination of chemical and physical mechanisms. Therefore, fossil remains found exposed on the surface are usually colonized by epilithic and endolithic lichens. The objective of this work is to determine the biodeterioration generated by lichens on fossil remains and its taphonomic implications. We identified the presence of the euendolithic lichens Lecidea andersonii and Athallia holocarpa growing into fossil penguin bones from Antarctica. The bioerosive damage was evaluated using light and electron microscopic techniques. Pits corresponding to apothecium and sinuous thin fissures remodeled, or in some cases produced, by hyphae were distinguished from the cracks originating from physical weathering. The maximum depth that hyphae extend inside the bone, probably constrained by the light supply, was established to be 2.5 mm. We provided a tool for the reconstruction of the chronology of the taphonomic events, describing the type and magnitude of the damage into the bones. Fil: García, Renato Andrés.
